Research Experience

(A). As a Ph.D. ScholarfromGLA University, Mathura and Central Institute for Research on Goats, Farah, Mathura (July, 2013 – Nov., 2019) (6 years 5 Month).

  • Cloning and expression of the secretary proteins (1693c, 2168c, Mod D, 85c, Pep AN, Pep AC) of MAP for development of an assay (ELISA) for differentiation of MAP infected and vaccinated animals (DIVA).
  • Immuno-proteomic analysis of recombinant secretory proteins has done (SDS-PAGE analysis, Protein estimation and Western blot analysis for Immuno-reactivity of proteins)
  • Recombinant CFPs based individual/cocktail ELISA have evaluated as a diagnostic marker with Indigenous ELISA kit in different domestic livestock species (Goats, sheep, cattle and buffaloes) is under process.
  • Optimization, laboratory scale and field validation of DIVA ELISA.
  • Species wise sero-reactivity and immuno-dominancy will be evaluated in individual secretory protein by western blot and plate ELISA and cocktail of secretory proteins will be evaluated by plate ELISA only.
  • Comparison of indigenous ELISA tests based on semi-purified protoplasmic antigens and recombinant secretary proteins for the diagnosis of spontaneous cases of Mycobacterium avium subspecies paratuberculosis infection in domestic ruminants and human beings.
  • Comparative study of c_ELISA, i_ELISA and Microscopy with respect to histo-pathological lesions in slaughtered and necropsied goats and slaughtered buffaloes naturally endemic for Mycobacterium avium subsp. paratuberculosis infection
  • Molecular diversity and homology in six CFPs genes in novel bio-type of Mycobacterium avium subspecies paratuberculosis of goat origin MAP ‘S 5’ Indian Bison type versus other strains
  • Cell mediated immunity and Rat spleenocytes cytotoxicity against six MAP CFPs.
  • Study of immunogenicity of CFPs in Immunized rabbits by c_ELISA, i_ELISA

(B) At Central Institute for Research on Goats, Farah, Mathura as a “Research Assistant” Since June, 2015 to June 2018 (3 years 1 Month) in ICMR project entitled “Crohn's disease in India: A multi-centric study from a country where Intestinal Tuberculosis as well as Johne’s Disease is endemic”.

  • Study the association of Mycobacterium avium subspecies paratuberculosis with Crohn’s Disease
  • Study microbiome, transcriptome and immune interactions in Crohn’s disease and Johne’s disease with reference to MAP infection.
  • Genetic finger printing and comparative genomics of human and animal MAP isolates.
  • Correlation of disease behaviour and treatment outcomes inulcero-constrictive ileocolonic disease (intestinal tuberculosis and Crohn’s disease) with and without MAP infection

  • Sub-typing/ Bio-typing: MIRU-VNTR typing of Mycobacterium aviumsubspecies paratuberculosis isolates using VNTR X3, VNTR3, VNTR7, VNTR10, VNTR25, VNTR32, VNTR47, VNTR292, MIRU 1, MIRU 4, VNTR259, VNTR1067.
  • Screening of commercial milk and milk products by multiple diagnostic tests (microscopy, ELISA, PCR, culture, LAT, dotELISA, FAT).
  • New isolates of MAP from samples originating from different sources were identified and confirmed and their glycerol stocks were prepared.

(C). Central Institute for Research on Goats, Farah, Mathura since January, 2012 to Dec.2014 (as a Research Associate): 3 years on Mycobacterium avium subspecies paratuberculosis (MAP) 

  • Isolation and identification of MAP from clinical specimens by using techniques such as Micoscopy, culture, PCR, qRT-PCR, ELISA, Immunofluorescence, LTT, LAT, FAT, Dot-ELISA etc.
  • Preparation of Johne’s disease vaccine, Vaccination, and evaluation of antibody and cell mediated immune response against MAP in animals (sheep, goats, cattle, buffaloes).
